Cloxacillin

Cloxacillin is an isoxazolyl penicillin. Penicillin is derived directly or indirectly from strains of fungi of the genus Penicillium and other soil inhabiting fungi grown on special culture media. The structure of penicillin consists of a thiazolidine ring connected to a β lactam ring, which is attached to a side chain. its use is limited to treatment of penicillinase-producing Staphylococcus aureus infections. Antibiotics require constant drug level in body for therapeutic effect. This is achieved by taking the drug at regular interval of time throughout the day and night as prescribed. Cloxacillin (Na) is important to take the drug for the full time period as prescribed. If you discontinue the drug it may result in ineffective treatment.

Characteristics
Cloxacillin [Na] is the derivative of Cloxacillin. It is of Semi Synthetic origin and belongs to Penicillin. It belongs to Peptidoglycan synthesis inhibitor pharmacological group on the basis of mechanism of action and also classified in Antibiotics pharmacological group.The Molecular Weight of Cloxacillin is 475.90. Its pKa is 2.7.
Contraindications
Cloxacillin is contraindicated in conditions like Hypersensitivity.
Effects
The severe or irreversible adverse effects of Cloxacillin, which give rise to further complications include Cholestatic jaundice, Hemolytic anemia, Blood dyscrasias, Seizures, Interstitial nephritis, Cholestasis, Neurotoxicity.Cloxacillin produces potentially life-threatening effects which include Pseudomembranous colitis, Anaphylaxis. which are responsible for the discontinuation of Cloxacillin therapy.The signs and symptoms that are produced after the acute overdosage of Cloxacillin include Vomiting, Diarrhea, GI disturbances, Abdominal discomfort.The symptomatic adverse reactions produced by Cloxacillin are more or less tolerable and if they become severe, they can be treated symptomatically, these include Skin reactions, Rashes, Phlebitis, Glossitis, StomatitisX, Nausea and vomiting, GI disturbance, Hyperthermia.
Indications
Cloxacillin is primarily indicated in conditions like Bacterial infections, Bone and joint infection, Cystitis, Endocarditis, Enterocolitis, Meningitis associated with spina bifida, Respiratory tract infections, Septicaemia, Skin infections, Soft tissue infections.
Interactions
Cloxacillin is known to interact with other drugs, the details of drug interactions is as follows:DrugDetailsSeverityOnsetManagementChloramphenicolChloramphenicol inhibit the effect of cloxacillin by inhibiting cellular protein synthesis and change bacteria into static one thus reducing the effectiveness of carbenicillin.ModerateCombination must be avoided. If necessary then cloxacillin administered several hours before chloramphenicol and closely observed.ProbenecidTetracycline (HCl) These interactions are sometimes beneficial and sometimes may pose threats to life. Always consult your physician for the change of dose regimen or an alternative drug of choice that may strictly be required.
Interfrence
Intereferes with Bradshaws Test for Bence-Jones proteins. Measurement of Urinary 17-oxo-steroids and 17-oxogenic steroids by MRC Method
Risks
Drug should not be given to patients suffering from Kidney dysfunction, patients suffering from Liver Malfunction, and Neonates.If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.
Storage
Caps, Susp, Inj Store in a well closed container, at room temperature.
Warnings
Cloxacillin should be used with caution in patients with any other illnesses or any allergy especially to penicillin or other antibiotics.This medication should be used only if clearly needed during pregnancy or lactation.
